    Drug dependency and drug use behaviour among male illicit drug users in selected rehabilitation centres in Sri Lanka

    Background: Illicit drug use has become a growing public health phenomenon in Sri Lanka. Many illicit drug users end up with drug dependency, becoming a burden to the country.Objectives: To describe drug dependency and drug use behaviour among male illicit drug users in selected rehabilitation centres in Sri Lanka.Methods: A cross-sectional study was conducted among 431 male illicit drug users in five selected rehabilitation centres in Galle, Colombo, Gampaha and Kandy districts, Sri Lanka. An interviewer-administered questionnaire was used for data collection. Drug dependency was identified using ICD symptom checklist for substance use disorder.Results: Majority of the sample were Sinhala (82.4%) and Buddhists (70.5%). Mean age (SD) was 32 (10.3) years. The highest percentage of the drug users were from Western province (75.6%). Of all illicit drug users, 97% were drug dependents according to ICD 10 symptom checklist. Heroin was most commonly addicted illicit drug (86.5%). Cannabis addiction was identified among 4.3% and all of them were from rural areas. Approximately 4% were addicted to multiple drugs. Injecting drug practices were reported among 23.4%. Mobile transfer of funds (eZ cash) was a popular method used to purchase drugs by drug users (21.8%) outside the Western province. In a shortage of the regular drug, 32.9% of drug users used alternative illicit drug, 3.2% used alcohol and 16.9% had a period of abstinence. Interestingly, 46.9% never experienced a shortage of drugs. Drug overdose was experienced by 25.1% of drug users.Conclusions: Drug dependency is a significant problem among illicit drug users. A proper system for early identification of drug dependents at community level is required to deliver better preventive care

    Metal transfer to sediments, invertebrates and fish following waterborne exposure to silver nitrate or silver sulfide nanoparticles in an indoor stream mesocosm.

    The fate of engineered nanomaterials in ecosystems is unclear. An aquatic stream mesocosm was explored the fate and bioaccumulation of silver sulfide nanoparticles (Ag2S NPs) compared to silver nitrate (AgNO3). The aims were to determine the total Ag in water, sediment and biota, and to evaluate the bioavailable fractions of silver in the sediment using a serial extraction method. The total Ag in the water column from a nominal daily dose of 10 μg L-1 of Ag for the AgNO3 or Ag2S NP treatments reached a plateau of around 13 and 12 μg L-1, respectively, by the end of the study. Similarly, the sediment of both Ag-treatments reached ~380 μg Ag kg-1, and with most of it being acid-extractable/labile. The biota accumulated 4-59 μg Ag g-1 dw, depending on the type of Ag-treatment and organism. The oligochaete worm, Lumbriculus variegatus, accumulated Ag from the Ag2S exposure over time, which was similar to the AgNO3 treatment by the end of the experiment. The planarian, Girardia tigrina, and the chironomid larva, Chironomus riparius, showed much higher Ag concentrations than the oligochaete worms; and with a clearer time-dependent statistically significant Ag accumulation relative to the untreated controls. For the pulmonated snail, Physa acuta, bioaccumulation of Ag from AgNO3 and Ag2S NP exposures was observed, but was lower from the nano treatment. The AgNO3 exposure caused appreciable Ag accumulation in the water flea, Daphnia magna, but accumulation was higher in the Ag2S NP treatment (reaching 59 μg g-1 dw). In the rainbow trout, Oncorhynchus mykiss, AgNO3, but not Ag2S NPs, caused total Ag concentrations to increase in the tissues. Overall, the study showed transfer of total Ag from the water column to the sediment, and Ag bioaccumulation in the biota, with Ag from Ag2S NP exposure generally being less bioavailable than that from AgNO3

    Effective delivery of hydrophobic drugs to breast (MCF-7) and Liver (HepG2) cancer cells: A detailed investigation using Cytotoxicity assays, fluorescence imaging and flow cytometry

    This study aimed to develop a drug carrier system consisting of a polymer containing hydroxyapatite (HAp) shell and a magnetic core of iron oxide nanoparticles. Doxorubicin and/or curcumin were loaded into the carrier via a simple diffusion deposition approach, with encapsulation efficiencies (EE) for curcumin and doxorubicin of 93.03 ± 0.3% and 97.37 ± 0.12% respectively. The co-loading of curcumin and doxorubicin led to a total EE of 76.02 ± 0.48%. Release studies were carried out at pH 7.4 and 5.3, and revealed higher release was at pH 5.3 expressing the potential application in tumor microenvironments. Cytotoxicity assays, fluorescence imaging and flow cytometry showed the formulations could effectively inhibit the growth of MCF-7 and HEpG2 cancer cells, being more potent than the free drug molecules both in dose and time dependent manner. Additionally, hemolysis tests and cytotoxicity evaluations determined the drug-loaded carriers to be non-toxic towards non-cancerous cells. These formulations thus have great potential in the development of new cancer therapeutics

    International audiencePhenotypic plasticity is an important but often ignored ability that enables organisms, within species-specific physiological limits, to respond to gradual or sudden extrinsic changes in their environment. In the marine realm, the early ontogeny of decapod crustaceans is among the best known examples to demonstrate a temperature-dependent phenotypic response. Here, we present morphometric results of larvae of the hairy crab , the embryonic development of which took place at different temperatures at two different sites (Antofagasta, 23°45′ S; Puerto Montt, 41°44′ S) along the Chilean Coast. Zoea I larvae from Puerto Montt were significantly larger than those from Antofagasta, when considering embryonic development at the same temperature. Larvae from Puerto Montt reared at 12 and 16°C did not differ morphometrically, but sizes of larvae from Antofagasta kept at 16 and 20°C did, being larger at the colder temperature. Zoea II larvae reared in Antofagasta at three temperatures (16, 20, and 24°C) showed the same pattern, with larger larvae at colder temperatures. Furthermore, larvae reared at 24°C, showed deformations, suggesting that 24°C, which coincides with temperatures found during strong EL Niño events, is indicative of the upper larval thermal tolerance limit.   is exposed to a wide temperature range across its distribution range of about 40° of latitude. Phenotypic plasticity in larval offspring does furthermore enable this species to locally respond to the inter-decadal warming induced by El Niño. Morphological plasticity in this species does support previously reported energetic trade-offs with temperature throughout early ontogeny of this species, indicating that plasticity may be a key to a species' success to occupy a wide distribution range and/or to thrive under highly variable habitat conditions

This system has a high ability in removing nutrients compared to conventional methods because it is a natural and environmentally friendly system (aquaponics). The current chapter aimed to review the possible application of aquaponics system to treat fresh market wastewater with the intention to highlight the mechanism of phytoremediation occurs in aquaponic system.     Serum biomarkers associated with SARS-CoV-2 severity

    Immunity with SARS-CoV-2 infection during the acute phase is not sufficiently well understood to differentiate mild from severe cases and identify prognostic markers. We evaluated the immune response profile using a total of 71 biomarkers in sera from patients with SARS-CoV-2 infection, confirmed by RT-PCR and controls. We correlated biological marker levels with negative control (C) asymptomatic (A), nonhospitalized (mild cases-M), and hospitalized (severe cases-S) groups. Among angiogenesis markers, we identified biomarkers that were more frequently elevated in severe cases when compared to the other groups (C, A, and M). Among cardiovascular diseases, there were biomarkers with differences between the groups, with D-dimer, GDF-15, and sICAM-1 higher in the S group. The levels of the biomarkers Myoglobin and P-Selectin were lower among patients in group M compared to those in groups S and A. Important differences in cytokines and chemokines according to the clinical course were identified. Severe cases presented altered levels when compared to group C. This study helps to characterize biological markers related to angiogenesis, growth factors, heart disease, and cytokine/chemokine production in individuals infected with SARS-CoV-2, offering prognostic signatures and a basis for understanding the biological factors in disease severity

    Effect of floor type on the performance, physiological and behavioural responses of finishing beef steers

    peer-reviewedBackground:The study objective was to investigate the effect of bare concrete slats (Control), two types of mats [(Easyfix mats (mat 1) and Irish Custom Extruder mats (mat 2)] fitted on top of concrete slats, and wood-chip to simulate deep bedding (wood-chip placed on top of a plastic membrane overlying the concrete slats) on performance, physiological and behavioral responses of finishing beef steers. One-hundred and forty-four finishing steers (503 kg; standard deviation 51.8 kg) were randomly assigned according to their breed (124 Continental cross and 20 Holstein–Friesian) and body weight to one of four treatments for 148 days. All steers were subjected to the same weighing, blood sampling (jugular venipuncture), dirt and hoof scoring pre study (day 0) and on days 23, 45, 65, 86, 107, 128 and 148 of the study. Cameras were fitted over each pen for 72 h recording over five periods and subsequent 10 min sampling scans were analysed. Results: Live weight gain and carcass characteristics were similar among treatments. The number of lesions on the hooves of the animals was greater (P < 0.05) on mats 1 and 2 and wood-chip treatments compared with the animals on the slats. Dirt scores were similar for the mat and slat treatments while the wood-chip treatment had greater dirt scores. Animals housed on either slats or wood-chip had similar lying times. The percent of animals lying was greater for animals housed on mat 1 and mat 2 compared with those housed on concrete slats and wood chips. Physiological variables showed no significant difference among treatments. Conclusions: In this exploratory study, the performance or welfare of steers was not adversely affected by slats, differing mat types or wood-chip as underfoot material

    Combinações entre cultivares, ambientes, preparo e cobertura do solo em características agronômicas de alface.

    Objetivou-se identificar combinações entre cultivares, ambientes de cultivo e preparo e cobertura de solo capazes de melhorar o desempenho agronômico e aumentar a produtividade da cultura da alface em cultivo orgânico. A pesquisa foi conduzida na Universidade Federal do Acre, utilizando o delineamento experimental de blocos casualizados, com parcelas subdivididas para cada experimento (campo e casa de vegetação), com quatro repetições. Em cada experimento, três cultivares de alface (Simpson, Marisa e Vera), constituindo as sub-parcelas, foram sorteadas nas parcelas, representadas por quatro preparos e cobertura do solo (encanteiramento com cobertura de palha de arroz, polietileno prateado, solo descoberto e plantio direto). A produtividade comercial de alface foi de 12,3 t ha-1 em cultivo protegido e de 7,9 t ha-1 em campo. O cultivo protegido promoveu melhor desenvolvimento das plantas, caracterizado por maior massa da matéria fresca e seca da parte aérea, massa da matéria fresca comercial e melhor classificação comercial, além de promover bom desempenho agronômico e maior produtividade em qualquer um dos preparos de solo. As cultivares Simpson e Marisa apresentaram massa da matéria seca da parte aérea semelhante e superior à ‘Vera’, porém, o crescimento do caule da ‘Simpson’ foi elevado, caracterizando pendoamento precoce, fato que reduz sua qualidade comercial. As cultivares Marisa e Vera não alongaram o caule indicando serem tolerantes às condições ambientais de Rio Branco. A cobertura do solo com casca de arroz ou plástico prateado contribuiu para minimizar os efeitos climáticos prejudiciais ao cultivo da alface em campo. O plantio direto orgânico não diferiu do plantio em canteiro descoberto

    Magnetic Anisotropic Energy Gap and Strain Effect in Au Nanoparticles

    We report on the observation of the size effect of thermal magnetization in Au nanoparticles. The thermal deviation of the saturation magnetization departs substantially from that predicted by the Bloch T3/2-law, indicating the existence of magnetic anisotropic energy. The results may be understood using the uniaxial anisotropy Heisenberg model, in which the surface atoms give rise to polarized moments while the magnetic anisotropic energy decreases as the size of the Au nanoparticles is reduced. There is a significant maximum magnetic anisotropic energy found for the 6 nm Au nanoparticles, which is associated with the deviation of the lattice constant due to magnetocrystalline anisotropy
